Taking early - mature pear variety Xinli No. 7 as test material, the author studied the influences of basic culture medium, embryo age, hormone and low temperature pre -treatment on the plantlet regeneration directly from the cultivated young embryos in vitro. The results indicated that the embryo germination rate and cotyledon green - turning on MS medium were similar to those on White medium. However, the rooting rate and seedling - forming rate at the latter culture stage on MS medium were better than those on White medium. The optimum hormone concentration and medium combination was MS +2. 0 mg/L 6 - BA +0.1 mg/L NAA. The culture of young embryos with the age of more than 85 d was more easily successful. The pre - treatment by low temperature had greater promotive effects on embryo germination and seedling - forming of 65 - day - old and 75 - day - old embryos than those of 85 - day - old embryos, and the improving effects of low temperature pre - treatment on the embryo germination and cotyledon green - turning were greater than those on the rooting rate and seedling - forming rate.%以早熟梨新梨7号为试验材料,在离体条件下研究了基本培养基种类、激素配比、胚龄大小及低温预处理等因素对早熟梨幼胚离体培养的影响.结果表明:MS培养基诱导新梨7号胚萌发和子叶转绿方面与White培养基相似,而在培养后期促进生根和成苗率方面优于White培养基;适宜培养基激素浓度比例为2.0 mg/L6 -BA +0.1 mg/L NAA;以胚龄85 d以后的幼胚培养较易成功.低温预处理对促进65 d和75 d胚龄的幼胚萌发和成苗效果优于85 d以后梨胚,且低温处理对胚萌发、子叶转绿的促进作用大于对生根率和成苗率的促进作用.
