Purification and determination of antioxidant activity of stilbenoids from the hairy root cultures of peanut.

Antioxidant stilbenoids such as piceatannol and resveratrol have documented beneficial effects on human health such as platelet aggregation effects, anti-inflammatory activities, vasorelaxation effects, and anticancer activities. However, other stilbenoids such as arachidin-1 and arachidin-3 are not commercially available, resulting in little research focusing on their purification and biological activity. Peanut hairy roots recently have shown to produce stilbenoids. The focus of this study was to purify sufficient quantities of piceatannol, resveratrol, arachidin-1, and arachidin-3 from Arachis hypogaea hairy root cultures. Centrifugal Partition Chromatography (CPC) using a heptane:ethyl acetate:ethanol:water in a 4:5:3:3 (v/v ratio) solvent system, operating in the descending mode, was used to purify the compounds. The biological activity of purified resveratrol, arachidin-1, and arachidin-3 was determined using the Thiobarbituric Acid Reactive Substances (TBARS) assay. Resveratrol, arachidin-1, and arachidin-3 were separated to a purity of 97.11%, 96.99%, and 91.75%, respectively. Piceatannol was not purified via CPC due to the insufficient amounts produced by the A. hypogaea hairy root cultures. Pure, standard resveratrol, piceatannol, and arachidin-1 inhibited LDL oxidation at doses of 27, 14, and 7 muM, respectively. Pure, standard arachidin-3 did not inhibit LDL oxidation at the tested doses. CPC purified resveratrol, arachidin-1, and arachidin-3 inhibited LDL oxidation at doses of 14, 7, and 14 muM, respectively.
