unique model of in vivo closed chest pulmonary artery catheterization and fluorescent videomicroscopy was used to determine the mechanism by which Platelet Activating Factor (PAF) produces pulmonary edema and to test the effect of WEB 2086, a PAF receptor antagonist, in sepsis-induced lung injury. Hypothesis. PAF induces pulmonary edema by increasing endothelial permeability without changes in pulmonary pressure gradient. Methods. Pulmonary artery pressure (PAP), heart rate (HR), blood pressure (BP) and pulmonary microvascular permeability (PMP) were measured in anesthetized rats with either fluorescent videomicroscopy (FV) or Evans Blue Dye (EBD) techniques. Three series of experiments were done: (1) Intravenous PAF (iv PAF) (n = 20); (2) Topical lung PAF (tl PAF)
