During the last few years isolated perfusion becomes more important in the treatment of isolated tumours. New techniques are developed to apply isolated perfusion to the liver and lungs. Bio-analytical methods are necessary to determine concentrations of the administered compounds.; A gas chromatographic-mass spectrometric (GC-MS) method was developed for the determination of L-PAM and its hydrolysis products. TNF-α was determined with a commercial ELISA-kit. However ELISA-tests can produce false positive results by the occurrence of cross reactions of the TNF-α antibody with other proteins. Therefore experiments which can give an impulse to determine TNF-α by capillary electrophoresis (CE) in the future are described in this thesis.; Isolated liver perfusions in pigs with L-PAM, MMC and TNF-α learned us that during an isolated perfusion a 20 to 40-fold concentration advantage for the regional circuit in comparison to the systemic circuit was found.; Isolated lung perfusions with L-PAM are safe and effective for the treatment of lung metastases of adenocarcinoma, in the rat. L-PAM concentrations in the lungs about 50 times higher in the isolated lung perfusion group than in the intravenous group.; Different capillary electrophoretic techniques were evaluated to separate TNF-α from plasma proteins with CE. Qualitative analyses of TNF-α in plasma were performed with this method.
