This work addresses the effect of the replacement of ammine group(s) by labile ligands (e.g., In reference to the interaction between nucleic acids and Previously, the formation of metal-DNA adducts was inferred by hydrolysis or enzyme digestion followed by gel electrophoresis, chromatography characterizations. However, these procedures are time consuming, tedious, and considerably increase the error possibilities. For the direct separation of oligonucleotides by IPC there are limits in number of nucleotides. In this work, a new separation technique: direct separation of calf thymus DNA (CTDNA) and its cobalt adducts by ion pairing modified size exclusion chromatography (IP-SEC) on reversed phase columns was developed by combining the advantages of both ion pairing chromatography (IPC) and size exclusion chromatography (SEC). The separation mechanism was investigated by changing mobile phase conditions.
