摘要

Temperate plant species undergo a large suite of physiological and biochemical changes during the fall in order to survive winter freezing. Abscisic acid (ABA) increases during this time and is an important modulator of freezing tolerance. The goal in our laboratory is to study the role of ABA in the development of freezing tolerance in the plant cell suspension culture system, Bromus inermis Leyss. The first objective of this project was to determine the effect of exogenous ABA on the physiology and expression pattern of phosphoglucomutase (PGM) mRNA in Bromus inermis Leyss.; The second objective of this study was to isolate and characterize a full-length cDNA encoding cytosolic PGM from Bromus. Two PGM clones, designated pBG-PGM1 and pBG-PGM2 were isolated from a cDNA library made from mRNA of ABA-treated Bromus suspension culture cells, using a previously isolated Bromus EST. The pBG-PGMI was selected for further sequencing and characterization. (Abstract shortened by UMI.)
