The size and exon composition of mammary gland and liver ceruloplasmin mRNA were studied by Northern analysis and RT-PCR. For Northern analysis, a 1.3 kb portion of human ceruloplasmin was cloned to server as a probe. The Northern autoradiographs showed that ceruloplasmin mRNA of mammary gland was indistinguishable in size from that of liver. For RT-PCR, oligo dT primers were used for reverse transcription of total pig liver and mammary gland RNA. For the PCR reactions, specific primers covering exons 1--19 of the human ceruloplasmin gene were used. All the paired primers for exons 2--18 gave products of the expected size in the case of liver. Similar studies were carried out with cDNA from mammary gland, and products of the same apparent size were obtained. The results suggest that the ceruloplasmin mRNA expressed by mammary gland is the same as that expressed in liver.
