Breast cancer is a disease affecting thousands of women every year. A common treatment for this disease is with apoptosis inducing drugs, also know as chemotherapy. One of the most commonly used drugs is doxorubicin. Development of resistance to this drug is a major factor limiting successful treatment of this and other cancers. The development of doxorubicin resistance is not fully understood. In order to better understand the mechanism of the development of drug resistance, studies were carried out to compare the proteomes of drug susceptible and doxorubicin resistant cells.; In order to perform this investigation, a non-gel based strategy for quantitative comparative proteomics was optimized and implemented. This proteolytic 18O labeling method was successfully applied to protein fractions from the two cell types. This study yielded multiple proteins that were up regulated in the doxorubicin resistant line.
