The effects of feeding different concentrations of curcumin on melanoma in mice were investigated. Sixty 4-week-old, female C57BL6 (Harlan) mice were randomly divided into four groups. Group 1 (control) was fed AIN-93G diet, while groups 2-4 (experimental) were fed 0.2% curcumin, 0.5% curcumin diet and 1% dietary curcumin throughout the study. On the 10th day, an aliquot (0.1 mL) of B16F10 cells suspended in RPMI 1640 (5 x 10 4 cells/mL) was injected subcutaneously into the right flank of each mouse. Four weeks later, mice were sacrificed and serum and tumors were collected. Sera were analyzed for VEGF concentration using ELISA immunoassay. The serum VEGF concentration of mice fed 1% curcumin (7.17 +/- 13.38 pg/dL) were lower than the controls (39.00 +/- 22.60 pg/dL) (p0.05). There was significant difference in average daily diet intake of mice in various groups, with highest intake in 1% curcumin group (2.88 +/- 0.11) (p0.05). There was no significant difference in tumor weight or tumor incidence between the groups. In summary curcumin, an established antitumor agent in-vitro lowered serum VEGF concentration but had no effect on tumor incidence or weight.
