Luman is a bZIP transcription factor belonging to the CREB/ATF superfamily and is believed to play a role in cell stress response and HSV-1 re-activation. c-Jun Activation Domain Binding Protein 1 (JAB1) has recently been identified as a putative binding partner for Luman by a yeast-two hybrid screening.;JAB1 appears to act as a transcriptional repressor for Luman by decreasing Luman's activation potential on GAL4-, CRE- and UPRE-responsive reporter genes. JAB1 does not appear to affect the cellular localization of Luman, but is likely involved in regulating its protein stability.;This study verified the interaction between Luman and JAB1 by four methods: (1) GST-pulldown, (2) immunofluorescence, (3) co-immunoprecipitation and (4) mammalian two-hybrid experiment. Furthermore, the MPN domain of JAB1 was shown to be necessary and sufficient for this interaction.
