Effectiveness of the cleaning and sanitation procedures was verified in seven supermarkets and five bakeries in Mayaguez, Rincon and Hormigueros towns. Microbiological tests performed included aerobic plate count (APC), coliform's total count, Escherichia coli, and ATP Bioluminescence (non-microbiological) tests. Samples were taken from food contact surfaces (slicer and its table) and non-contact surfaces (environmental samples) from the refrigerator's door handle and the floor drainage in two time periods: before starting operations in the morning (AO) and after four hours of operations (DO). The statistical analysis results from the T test showed that there were no significant differences (P = 0.05) AO and DO in the direct contact and non-contact surfaces in supermarkets and bakeries for the microbiological analysis. In general, APC values demonstrated that the establishments did not meet with the standards for the studied surfaces. In the coliform test, Escherichia coli was found in 2 of 96 collected samples that infers that the personnel that works in these establishments complies with the personal good hygiene practices like washing their hands constantly. The application of ATP bioluminescence needs further studies in order to validate its usefulness as a verification tool for cleaning and sanitation by inspectors, supervisors and/or managers. No relationship was found between colony forming units (CFU) and relative light units (RLU). The study demonstrated the need to develop and implement standard sanitary operating procedures (SSOP's) and monitoring and verification procedures at retail and deli establishments.
