By the experiment of quadratic orthogonal rotation combination design, the influence of different assay conditions on the activity of chalcone isomerase (Y) in Ginkgo biloba has been probed into in this paper, they include reaction temperature (X1), pH value (X2), reaction time (X3), substrate concentration (X4). The mathematical model of the CHI activity has been established which are influenced by these determination conditions: Y = 55.6167-0.7283X1-1.7258X2-0.3633X3-0.6217X4-2.3521X12-1.7908X22-0.9783X32-0.7121X42-0.0125X1X2-0.0088X1X3-0.0800X1X4+0.3238X2X3-0.1225X2X4-0.0813X3X4. It is concluded from the model that when temperature, pH value, time and substrate concentration are: 35 °C, 6.8, 40 min and 20μM respectively, the yield (Y) is the maximal (55.62 U/g) and consistent with the validate experiment result.
