The study is aimed at solving a significant challenge of extending the half-life of therapeutic proteins using crystalline biopharmaceuticals and without redesigning the molecules. Recombinant human interferon α-2b (rhIFN) was crystallized by chelating with zinc in the presence of protamine. The resulted rhiIFN crystal formulations containing protamine exhibited good sustainedrelease potential both in vitro and in vivo. rhIFN well kept their structural and functional integrity in these formulations. The approach described here offers an exciting possibility of being broadly applicable to other therapeutic proteins.
