Preparation and in vivo / in vitro evaluation of a tri-layer tablet containing gimepiride (IR)/metformin (SR) with an isolation layer

摘要

Purpose: The objective of the present study was to develop a tri-layer tablet containing gimepiride (IR)/metformin (SR) with an isolation layer which was administered once daily to improve diabetes patients compliance and achieve better glycemic control. Methods: IR (immediate-release) layer, isolation layer and SR (sustained-release) layer were all prepared by wet granulation technique and the trilayer tablets were prepared by trebly compression technology. Results: It was found that the stability of gimepiride was elevated obviously. Metformin hydrochloride in the preparation had sustainedrelease character obviously and the in vivo-in vitro correlation was obvious. Conclusion:The present study successfully prepared a tri-layer tablet which had both IR and SR characters. The preparation is effective in glycemic control rapidly and persistently
