Immunostimulatory CpG ODN delivery with β-1,3-glucan receptor of Dectin-1

Unmethylated CpG sequences (CpG ODN) stimulate Toll-like receptor (TLR9) in antigen presenting cells (APCs) such as macrophages, and evoke T helper 1 (Th1) phenotype by secreting various cytokines. Recently, we have reported that a β-1,3-glucan polysaccharide, schizophyllan (SPG) forms a complex with CpG ODN, and can be applicable to CpG ODN carriers. Since SPG and other β-1,3-glucans bind to Dectin-1 (a C-type lectinlike pattern recognition receptor) expressed on cellular surface of APCs, the complex can get a selectivity for CpG ODN delivery into APCs. This study shows that SPG and its complex can be recognized by Dectin-1 on its transfectant. The result indicates that the complex can improve the activity of CpG ODN for the selective recognition.
