Oxidative stress plays an important role in Parkinson's disease (PD), suggesting that an antioxidant may be an ideal option for the treatment of PD. Schisanhenol (Sal), an active component from a Chinese herb, has significant antioxidant activity. In this study, we investigated the effect of Sal against oxidative stress induced by 1-methyl-4-phenylpyridinium ion (MPP~+) in human neuroblastoma SH-SY5Y cells. MPP~+-induced neuronal injury was characterized by the decrease of glutathione (GSH), glutathione peroxidase (GSH-Px), catalase (CAT), superoxide dismutase (SOD), increase of malondialdehyde (MDA) levels and the release of lactate dehydrogenase (LDH). Pretreatment with Sal markedly attenuated MPP~+-induced oxidative stress, indicated by the increase of GSH, GSH-Px, CAT, SOD activities, decrease of MDA levels and LDH release in the biochemical assay. Flow cytometry analysis demonstrated that Sal inhibited the production of reactive oxygen species (ROS) caused by MPP~+. The above effects were in a dose-dependent manner. These results suggest that Sal has neuroprotective effects against MPP~+-induced oxidative stress in SH-SY5Y cells, which might be a potential therapeutic agent for preventing or treating PD.
