To better understand the osmo-adaption mechanism in the Torulopsis versatilis, we investigated the salt-tolerant gene HOGl from the wild-type T. versatilis. Genome of T yeast was used for PCR reaction to amplify the HOG1 gene. The PCR productions were sent into sequencing vector pUC19 to achieve the plasmid pUC19-HOG1. The engineered strain W303-Yep195-T and the control strain W303-YEp195 were constructed, respectively. It was found that overexpression of THOG1 could obviously increase the tolerance of salt, compared to the control. Identification and overexpression of THOG1 play an important role for the osmo-adaption mechanism of the T. versatilis used in soy-sauce fermentation.
