Myelin formation in the peripheral nervous system is dependant on axonal signaling to Schwann cells, which generate the myelin sheath. Activation of the NF-kB transcription factor in Schwann cells is required for initiating myelin formation in these cells. We report that the axonal membrane bound ligand, Neuregulin 1 (NRG1) type III, is sufficient for the activation of NF-kB in Schwann cells. This activation is mediated by ErbB2 and is required for NF-kB activity in myelinating cells both in vitro and in vivo.
