Clinical issue for developing mucosal vaccine and the solution how to meet with them were discussed based on the results of animal experiments. Flt3 ligand and CpG enhanced antigen-specific IgA responses in external secretions for a long period, indicating strong and long acting adjuvant effect. PC-specific IgA in nasal wash and IgG in serum induced by intranasal immunization with PC together with CT were cross-reactive with all strains of S. pneumoniae and H. influenzae. Further, intranasal immunization with PC did not increase serum IgE. Sublingual immunization with PC also induced mucosal as well as systemic immune responses and did not increase serum IgE level. Those results suggest PC is one of the candidates of broad spectrum mucosal vaccine and sublingual vaccination with PC might be useful to prevent acute otitis media.
