Deoxynivalenol (DON) is produced mainly by Fusarium graminearum and F. culmorum which frequently contaminate raw and processed cereal grains and animal feeds, which has caused outbreaks of poisoning in humans and animals. Extrusion processing may provide a crucial detoxification strategy to reduce or eliminate toxicity of DON. The objective of this study was to determine loss of toxicity of DON in extruded corn grits by the MTT bioassay using Chinese hamster ovary cell line (CHO-K1), and compare theresults to chemical (HPLC) and biochemical (ELISA) methods of analyses. A split-split plot design was used for the extrusion process experiments at temperatures of 150, 175, and 200 and screw speeds of 70, and 140 rpm. The effect of extrusion temperatures on loss of DON was found to be linear. The percent reductions of DON ranged from 2226%, 2628%, and 3135% at temperatures of 150, 175 and 200, and screw speeds of 70 and 140 rpm, respectively, as measured by HPLC analysis. The MTT bioassay results weremore closely correlated with HPLC results (correlation coefficient, r = 0.90) than ELISA results (r = 0.78). The MTT bioassay, using a sensitive mammalian cell line, was proven to be a useful method for quantification of DON, as well as a potential screening method for toxicity testing of contaminated extruded cereal based products.
