Proteomic analysis of plasma samples by LC-MS has become a mainstay for biomarker development. A major barrier to comprehensive analyses of plasma proteins is the presence of several highly-abundant proteins. Numerous approaches have been developed to deplete these proteins in order to achieve greater sequencing depth. These techniques typically involve selective precipitation of plasma proteins or chromatographic depletion of specific highly-abundant proteins. Here, we present a novel approach for plasma fractionation that involves selective re-solubilization of plasma proteins with hexafluoroisopropanol (HFIPA) following a methanolic precipitation of crude material and compare it to an established protocol.
