1. Pag+ Y. pestis can palmitoylate lipid IV_A. 2. PagP+ Y. pestis adds 2 C16 residues in a novel manner. 3. PagP+ Y. pestis LPS grown at 37°C is significantly more immunostimulatory than sWT Y_p LOS grown at the same temperature. 4. PagP+ Y. pestis LPS grown at 37°C can activate the TRIF/TRAM arms of the TLR4 signaling pathway. 5. In-vivo studies in mice (BALB/cJ or mice with human TLR4-MD2 receptors) showed no difference in virulence between the PagP repaired or wild type Yp strains (KIM or CO92). 6. PagP+ Y. pestis LPS grown at 37°C has the ability to stimulate the Innate Immune system and would make it an ideal candidate for developing a new adjuvant.
