Solvent conditions for MBP isolation were optimized, with propanol and ethanol at pH <2 yielding high total MBP and consistent splice variant and PTM ratios LC-MS successfully separated dominant splice variants and PTMs and allowed for observation of potential unknown MBP variants Small but statistically significant variation in splice variant ratios in brain vs spine were observed.
