DNA stable isotope probing (DNA-SIP) with ~(13)C-labeled substrates can be an effective method for identifying microorganisms that degrade and assimilate a particular contaminant, but there are some challenging limitations to this approach. These include difficulties in dealing with contaminant mixtures or aged contaminants in soils. We tested a SIP approach that, if successful, could facilitate understanding the degradation of multiple or aged pollutants in soil. We specifically hypothesized that DNA-SIP with H_2~(18)O could be used to characterize pollutant degrading microorganisms in soil.
