Abstract: Studies on the complementary mating types of Parameciumprimaurelia (Protozoa, Ciliates) have shown that celllines which differ from each other in mating typeexpression are characterized by different cellcontents, organization, and physiology. Referring tothese differences and to the differential rates of foodvacuole formation, oral apparatuses of the two matingtype cells are assumed to possibly differ from eachother in some traits, such as, for instance, in theirlengths. In our work, the highly organized oralstructures are analyzed by means of a laser scanningconfocal optical microscope (CLSM), which providestheir 3-D visualization and measurement. The extractionof the 3-D intrinsic information related to thebiological objects under investigation can be in turnrelated to their functional state, according to theclassical paradigm of structure to functionrelationships identification. In our experiments, weacquired different data sets. These are optical slicesof the biological sample under investigation, acquiredin a confocal situation, through epi-illumination, inreflection, and, for comparison with conventionalmicroscopy, 2-D images acquired via a standard TVcamera coupled to the microscope itself. Our CLSMsystem is equipped with a laser beam at 488 and 514 nmand the data have been acquired with various steps ofoptical slicing, ranging from .04 to .25 micrometers.The volumes obtained by piling-up the slices arerendered through different techniques, some of themdirectly implemented on the workstation controlling theCLSM system, some of them on a SUN SPARC station 1,where the original data were transferred via anEthernet link. In this last instance, original softwarehas been developed for the visualization and animationof the 3-D structures, running under UNIX and X-Window,according to a ray-tracing algorithm. !81
