Demineralized bone matrix proteins are known to induce bone formation. Several bone morphogenetic proteinshave also been shown to produce bone in heterotopic locations which would limit the use of the compound forbone augmentation. The objective of this investigation was to evaluate the tissue-implant response in the fibroustissue surrounding tricalcium phosphate (TCP) implants containing demineralized bone matrix proteins whenimplanted subcutaneously. Sixteen animals in two experimental groups (n = 8/group) were implanted with either aTCP implant or a TCP implant containing demineralized bone matrix proteins. At 30 days post-implantation, thefibrous tissue surrounding the implants was harvested. Sections of stained fibrous tissue were evaluated for thepresence of macrophages, bone, fibrocytes, neutrophils, vascularity and thickness using semi-automatedquantitative methods. Tricalcium phosphate delivery systems are capable of delivering demineralized bone matrixproteins subcutaneously over 4 weeks with minimal evidence of ectopic bone formation.
