Cloning and Expression Analysis of MdFLC Gene Involved in Floral Development in Malus Domestica

Based on the results of previous study on columnar apple using RNA-seq technology,one cDNA sequences of homologous gene ofMADS-box gene family from columar apple was cloned by RT-PCR,which was named as MdFLC.The MdFLC contains a 603 bp open reading flame (ORF) corresponding to a deduced protein of 201 amino acids,while the estimated molecular weight and isoelectric point (pI) of the putative protein were 47.68kD and 5.18.The results indicated that MdFLC had 1 to 75 amino acids highly conserved MADS-box domain,105 to 175 for the K-box domain.The phylogenetic tree of FLC showed that MdFLC was closest with sand pear FLC and had 94.50% the identity in protein,followed by hickory nut FLC,and the evolutionary relationships of homologous genes in cabbage and radish were the farthest.The real-time quantitative PCR analysis showed that MdFLC gene was always expressed in flower bud and different parts of flower both in columnar and standard apple trees.The MdFLC gene expression on the flower buds and the parts of the flowers (Petal,Pistil/Stamen,Sepal,Receptacle) of columnar apple trees of early flowering lines (95-31,95-41,95-45,95-107,95-152) were significantly lower than that of Fuji same position.But the expressions of MdFLC gene have no obvious regularity between columnar apple trees of early flowering lines.
