Intradermal immunization is a potentiallypowerful technology, which can enhance immuneresponses and provide antigen sparing. It can berealized through the use of microneedles, whichcontain solid state vaccine formulations dissolvableupon administration to the skin. The technology,however, is not fully compatible with many existingvaccine adjuvants. A new approach is suggested, inwhich a well-defined macromolecular compound,poly[di(carboxylatophenoxy)phosphazene] (PCPP),is employed both as an immunoadjuvant and as amicrofabrication polymer. Dual functionality of thispolymeric excipient can potentially simplify andimprove the efficacy of microneedle vaccine deliverytechnology. Testing of PCPP, in vivo and in vitro,has shown that it is compatible with solid statemicroneedle methods and demonstrated that PCPPis potent intradermal adjuvant.
