Sito-specific PEGylation of G-CSF by reversible denaturation

摘要

Granulocyte-colony stimulating factor (G-CSF)was conjugated to a single chain of poly(ethylene)glycol in order to obtain a sito-specific product. Thepolymer was bound to the lone thiol group of Cys17,after a partial denaturation of the protein. The conjugateobtained after renaturation was assessed by CD spectraand the biological activity evaluated by a cell cultureassay was close to the starting product.
