A novel soybean seed ferritin gene, SferH-5, was firstly isolated from 7-day-old soybean seedlings. We speculate that SferH-5 encodes a novel 26. 5-kDa subunit of soybean seed ferritin, which is designated H-5. After the target fragment was confirmed by DNA sequencing, the gene sferH-2 and sferH-5 were correctly inserted into the prokaryotic expression plasmids vector pET28a and the yield recombinant plasmid was transformed to the E. coli BL21 (DE3). H-2/H-5 complex was successfully expressed in E. coli and collected by affinity chromatography with Ni2+ column.
