A new gene carrier, water soluble lipopolymer (WSLP), was synthesized and evaluated as a carrier of IL-4 expression plasmid, in which the expression of IL-4 was driven by rat insulin promoter. Gel retardation assay confirmed that WSLP formed complex with pRIP-IL4. In addition, DNase I protection assay proved that WSLP protected plasmid DNA from DNase I. WSLP had less toxicity to MIN6 cells than PEL WSLP/pRJP-IL-4 complex showed tissue specific and glucose responsive expression of IL-4 in mouse insulinoma cells. This gene delivery system is useful for the development of gene therapy method for the treatment or prevention of diabetes.
