Extracts from mycelia of G. lucidum were studied for their growth inhibitory activity towards cancer cell lines. Hot water extract (WE) of G. lucidum was first prepared. Triterpene-enriched fraction as ethanol soluble portion of WE (WEES) was obtained by removing the ethanol insoluble polysaccharide precipitates from WE. Fraction WEES was found to inhibit growth of some cancer cell lines. WEES was further separated by Sephadex G-25 into ten fractions and the sixth fraction, WEES-G6, was found to possess the highest growth inhibitory activity. WEES-G6 exerted growth inhibitory activity on human hepatoma cell line, Huh7, but only had little influence on normal liver cell line, Chang liver. Further molecular analysis showed that WEES-G6 caused obvious decreases in the activity of SOD and the level of sulfhydryl contents as well as an increase in the level of H<,2>O<,2> in Huh7 cells. On the other hand, these changes were not shown by Chang liver cells. The oxidative stress induced by WEES-G6 could be one of the reasons accounting for growth inhibition in Huh7 cells.
