Isolation and Identification of Bacillus pumilus KF1 with Novel Thermoalkalotolerant Cellulases

A cellulase-producing strain, designated KF1, was isolated from compost. Cells were Gram-positive, rodshaped and spore forming. Strain KF1 was identified as a strain of Bacillus pumilus based on phenotypic, physiological and biochemical characteristics, as well as 16S rDNA phylogenetic analysis. The maximum carboxymethyl cellulase (CMCase), filter paper cellulase (FPase) and P-glucosidase activities reached 200, 245, and 225 U/mL after 60, 72 and 84 h of cultivation, respectively. Maximum CMCase activity was measured at pH 6.0 and 60℃. It had both good alkaline endurance and stable enzymatic activity at pH value 5.0-9.0 and had remarkable heat stability under 65 ℃ as well. These enzyme properties are attractive for potential application in industries.
