Mammalian cells have been widely used as hosts for producing recombinant therapeutic proteins due to their capacity for post-translational processing,particularly glycosylation necessary for the effectiveness of therapeutic proteins in vivo. The majority of proteins of pharmaceutical interest are glycoproteins, the carbohydrate part of which plays important biological roles: immunogenicity,solubility, recognition, protection from proteolytic attack. In the process of cell culture, different environmental factors, including temperature, dissolved oxygen, pH,osmosality, and medium components so and so on, have significant influence on the glycosylation of recombinant proteins. The effects of different culture conditions on the glycosylation of recombinant proteins are reviewed in this paper. The characterization of effects of the critical environmental parameters on glycosylation should enable process control to improve glycosylation process.
