摘要

Bromelain enzyme (EC 3.4.22.4) is a protease of molar mass of 31 kDa with numerous applications in the food industry as well as in medicine and pharmacology, with specific properties which make this enzyme one of the best vegetal proteases. Membrane separation process is an alternative among or amongst other separation process, recovering and purifying, macromolecules. This work was study the bromelain enzyme recovery in plain membrane system by experimental factory planning of the type 2~2, with pH and pressure as factory and % yield as response. Protein concentration was measured during process time by Bradford method and specific enzymatic activity was determinate according to Murachi and Baldini method. Experimental data were possibility to make the adjust of the empiric model by ANOVA methodology and process optimization by RSM methodology. Fitting empiric model was of type y=b_0+b_1~*x_1+b_2*x_2+b_12*x_1*x_2 with multiple correlation approaching of one and response surface analysis showed optimal operation zone of membrane plain is that tend to low pressure.
