Introduction MicroRNAs (miRNAs) are a class of small non-coding RNAs that regulate the expression of target genes at the post-transcriptional level by transcript degradation or translational inhibition.They are involved in the regulation of many important physiological processes (Lewis et al.,2005).Some studies showed that miRNAs were involved in the regulation of mammary gland (Wang and Li,2007),the organ that can undergo cycles of cell division,differentiation and dedifferentiation in the adult.The PRLR is a member of the cytokine receptor superfamily,associate with JAK2/STAT5 signaling pathway (Eric et al.,2010).Using TargetScan6.2 predict software,PRLR gene was predicted to be the potential target gene of miR-142-3p.So it is necessary to confirm the regulatory relationship between miR-142-3p and PRLR.In this study,miR-142-3p mimics or inhibitor were transfected into bovine mammary epithelial cells respectively.Using quantitative real-time PCR (qRT-PCR),western blotting,and CASY TT techniques the expression of lactation related genes and proteins was addressed and analyzed.
