Disclosed is an immunofluorescence kit for detecting the PD-L1 gene expression of a patient with an esophageal squamous cell carcinoma by means of peripheral blood circulating tumor cells. The kit mainly comprises a primary antibody suspension composed of goat serum, a mouse anti-CK gene, a rat anti-CD45 gene and a rabbit anti-PD-L1 gene, and a secondary antibody suspension composed of a fluorescently labeled goat anti-mouse antibody, a fluorescently labeled goat anti-rat antibody and a fluorescently labeled goat anti-rabbit antibody. The detection method mainly comprises: collecting peripheral blood, treating the peripheral blood, filtering and enriching circulating tumor cells, and detecting the PD-L1 gene expression of the circulating tumor cells by means of an immunofluorescence method.
