plasmid with nucleotide sequence - a sequence coding for a or b chain of insulin and still for its framstellning and e.coli plasmid - and still for framstellning of e.coli
New DNA transfer vector contains in its nucleotide sequence a sub-sequence having the structure of and transcribes from, a gene of a higher organism. Specifically the sub-sequence is the code for human insulin A or B chain (or both), or human growth hormone (the nucleotide sequences of these codes are given). Also new are microorganisms including in their nucleotide structure such as a sub-structure. - The microorganisms are made by first isolating an appropriate messenger-RNA; purifying it, then using it to elaborate a double-strand complementary DNA (c-DNA) contg. the required code. This is then reacted with a DNA-transfer vector having reactive end-groups able to react with itself or with the c-DNA. - The modified microorganisms can be used to produce insulin or growth hormone by fermentation, which is easier and quicker than cultivation of cell lines and gives better yields.
