PURPOSE: A mutant strain of Corynebacterium ammoniagenes capable of producing thymidine and a fermentation method of producing thymidine using the same strain are provided, therefore thymidine can be produced in higher yield. CONSTITUTION: The mutant strain of Corynebacterium ammoniagenes ThyM3(KCCM-10225) capable of producing thymidine is isolated by preparing a 5-fluorouracil tolerant strain (type 1) by treatment of wild Corynebacterium ammoniagenes(ATCC-6871) with NTG; preparing a hydroxyurea tolerant strain (type 2) by treatment of the strain (type 1) with NTG; preparing a trimethoprim tolerant strain (type 3) by treatment of the strain (type 2) with NTG; preparing a thymidine phosphorylase deficient strain (type 4) by treatment of the strain (type 3) with NTG; preparing an inosine requiring strain (type 5) by treatment of the strain (type 4) with NTG; preparing a 5-fluorocytosine tolerant strain (type 6) by treatment of the strain (type 5) with NTG; preparing a thymidine kinase deficient strain (type 7) by treatment of the strain (type 6) with NTG; and preparing a thymidine tolerant strain by treatment of the strain (type 6) with NTG.
