PURPOSE: A single dominant male sterile line and a restorer gene for male sterility line in broccoli and a method of producing the same are provided, which single dominant male sterile line is useful for producing a first generation hybrid broccoli having improved productivity. CONSTITUTION: The method for producing the single dominant male sterile line in broccoli comprises the steps of: treating fertile Brassica oleracea var. italica 5159 with a mutation inducing agent during its microspore culture to select dominant male sterile lines; hybridizing the dominant male sterile lines with pollens of fertile lines to obtain a F1 generation; and propagating the F1 generation by using cuttings of lateral shoot and hybridizing the F1 generation with the fertile lines to obtain a sterile line of F2 generation, wherein the mutation inducing agent is 0.01 to 0.1 microM of NMU(N-nitroso N-methyl urethane). The method for producing a restorer gene for male sterility line in broccoli comprises the steps of: treating fertile Brassica oleracea var. italica 5159 with a mutation inducing agent during its microspore culture to select dominant male sterile lines; hybridizing the dominant male sterile lines with pollens of fertile lines to obtain a F1 generation; propagating the F1 generation by using cuttings of lateral shoot and hybridizing the F1 generation with the fertile lines to obtain a sterile line of F2 generation; and continuously hybridizing the sterile line of F2 generation with fertile lines to obtain a F5 generation containing the restorer gene for male sterility line in broccoli.
