Recombinant constructs, vectors and expression cassettes are provided herein which include a first promoter which is suitably a trna promoter operably linked to a first polynucleotide encoding a first individual guide rna and a second promoter operably linked to a second polynucleotide which encodes a cas9 polypeptide. the first individual guide includes a first strand complementary portion of a DNA target sequence and a second portion capable of interacting with the cas9 polypeptide. Also provided are cas9 polynucleotides and polypeptides derived from codon optimized staphylococcus aureus with nuclear localization signals and, optionally, an epitope tag. Constructs for the production of sgrnas are also provided, including a trna. Viral replication inhibition methods, inhibition of expression of a target sequence from a virus, or treatment of a viral infection or cancer by viral induction using the compositions are also provided.
