The present invention relates to a humanized death receptor (DR)-4 antibody gene with cell death-inducing activity and a dual-action chimeric antigen receptor T cell or a natural killer (NK) cell medicine using the same. According to the present invention, a secretory DR4 scFv antibody recombination protein synthesized by using humanized anti-DR4 scFv binds to DR4 expressed on the surface of a cancer cell to induce death of the cancer cell is to be used as a DR4-specific targeted anticancer agent. Also, a chimeric antigen receptor (CAR) using humanized anti-DR4 scFv can be used as a DR4-specific CAR-T cell or a CAR-NK cell medicine to bind to DR4 expressed on the surface of the cancer cell to expect a strong immunization anticancer effect as a dual-action cell medicine capable of simultaneously exhibiting cancer cell death induction by DR4 and cytotoxicity of a cytotoxic T lymphocyte or a natural killer cell. Also, since chemotherapy and radiotherapy used in cancer treatment occasionally increases DR4 expression of cancer cells, the DR4-specific CAR-T cell or CAR-NK cell can be used with existing anticancer drugs or radiotherapy to be used as a novel method of treating intractable cancer.
