APPLICATION OF EPALRESTAT IN PREPARATION OF PANCREATIC CANCER DRUG AND METHOD FOR VERIFYING INHIBITING EFFECT ON EXOSOME SECRETION OF PANCREATIC CANCER CELL

The present invention provides an application of epalrestat in preparation of a pancreatic cancer drug, and the pancreatic cancer drug is applied to inhibiting exosome secretion of a pancreatic cancer cell. A pharmaceutical composition for resisting pancreatic cancer contains epalrestat. The pharmaceutical composition is applied to inhibiting exosome secretion of the pancreatic cancer cell. Embodiments of the present invention further provide a method for verifying the inhibition effect of epalrestat on exosome secretion of the pancreatic cancer cell, comprising the following process: extracting a cell supernatant exosome by using a low-temperature ultracentrifugation method; quantifying protein by using a BCA kit after lysing the collected exosome, and reflecting the amount of the exosomes by using the measured amount of the protein; verifying a double-layer lipid membrane wrapped cup-shaped structure of the exosome by using a transmission electron microscope; and detecting exosome protein concentration by means of protein polyacrylamide gel electrophoresis having Coomassie brilliant blue. The present invention provides new use of epalrestat, i.e., the inhibition of exosome secretion. The epalrestat has great application potential in clinical tumor treatment.
