Interactions of Natural and Modified Collagens with Human Blood Platelets.

The report describes the quantitative determination of the actual amount of dispersed collagen which binds to platelets during the aggregation reaction in platelet-rich plasma. A titration proves that,under the usual conditions of the aggregation experiment,the reaction rate is not limited by the amount of collagen bound. The authors compare the binding to platelets of alpha 1 (I) and alpha 2 (I),chains,whole denatured Type I collagen from tendon. Observations of the interactions of Type I collagen with glucose,sucrose,and Ficoll (polymerized sucrose) are reported. In the presence of sucrose and Ficoll,collagen aggregation and adhesion to surfaces are greatly enhanced. Studies of binding of sucrose and glucose to collagen bydialysis and viscometry are described. In Appendix A the authors describe a new method for characterizing the kinetics of the platelet response to stimulating agents,by simultaneously and continuously monitoring aggregation and release of ATP.
