Comparison of Plasma Absorbance Changes Due to Stress and Acute Lung Injury

The purpose of this study was to determine whether non-specific stress induced spectrophotometric absorbance changes in rodent plasma and then to compare these results with spectrophotometric data from toxic gas-exposed rodents. The objective was to determine whether the increase in absorbance was phosgene (CG) specific. Five groups of rats and guinea pigs were used. They were (a) CG-exposed, (b)immobilized stress, (c) immobilized stress exposed to CG, (d) free-roaming controls and (e) perfluoroisobutylene.exposed. Gas-exposed groups were exposed to an already determined concentration of gas known to produce pulmonary edema. Stress groups were immobilized and deprived of food and water for one hour. Plasma was scanned in a spectrophotometer from 200 - 700 nm. The area under the curve was calculated between 375-450 nm. Differences between guinea pig groups were not reflected by plasma analysis. The rat data showed that there was a significant stress effect. Since stress immobilization did produce changes in plasma absorbance, plasma absorbance cannot be used to diagnose CG exposure.
