Enzymatic Release and Macrolactonization of Cryptophycins from a Safety-Catch Solid Support

摘要

Many natural products of pharmacological importance consist of macrocyclic structures,for example the polyketide antibiotic erythromycin,the non-ribosomal peptide cyclo-sporine,and the cryptophycins,a family of depsipeptides and potent antitumor agents.In nature,these macrocyclic compounds are synthesized by modular enzymatic "assembly lines" consisting of polyketide (PK) synthases,non-ribosomal peptide (NRP) synthetases,and hybrid NRP/PK synthetases.
