Because solubility and permeability form two of the three critical factors determining oral absorption (the third being first-pass clearance), careful treatment of electrolytes is critical for understanding the fate of an orally dosed compound as it passes through the gastrointestinal (GI) tract and eventually enters the bloodstream (1). While the blood is maintained at pH = 7.4 through homeostatic mechanisms, the pH along the GI tract varies greatly from stomach to colon.
