DEVELOPMENT OF ELISA TEST KITS FOR DETECTING ANTIBODIES OF PORCINE REPRODUCTIVE AND RESPIRATORY SYNDROME (PRRS)

MARC-145 and identified as the US strain (01NP1) and EU strain (01RB1). Gene coding for nucleocapsid (N) or ORF7 was isolated and amplified by RT-PCR. The PCR product was cloned into pBAD Directional TOPO~R Expression Vector and nucleocapsid recombinant proteins from each strain was expressed in the TOP10 Escherichia coli. The 6xHis-ORF7 recombinant proteins were purified in a single step by Ni-NTA Column affinity chromatography with approximated molecular mass of 30 kDa and were used for ELISA comparing to the commercial ELISA. The results from the recombinant proteins coated ELISA were acceptable.
