胃腺癌组织长链非编码RNA TUG1表达变化及其与p53蛋白的关系

摘要

目的 观察胃腺癌组织长链非编码RNA牛磺酸上调基因1(TUG1)表达变化,并分析其与p53蛋白的关系.方法 收集108例胃腺癌患者肿瘤组织及其癌旁正常组织,采用real-time PCR法检测各组织TUG1表达,采用免疫组化法检测各组织p53表达,分析胃腺癌组织TUG1表达与患者临床病理特征及p53表达的关系.结果 胃腺癌组织TUG1相对表达量为2.36±1.01、高表达90例,高于癌旁正常组织的0.93±0.54、18例(P均＜0.01).TUG1相对表达量在胃腺癌低分化者高于高分化者,浸润深度T3、T4者高于T1、T2者,有区域淋巴结转移者高于无区域淋巴结转移者,有远处转移者高于无远处转移者,临床分期高者高于临床分期低者(P均＜0.05),但不同年龄、性别、肿瘤直径及有无脉管侵犯、神经侵犯患者TUG1相对表达量比较差异均无统计学意义(P均>0.05).胃腺癌组织p53阳性表达率为61％,高于癌旁正常组织的39％(P＜0.01).Pearson相关分析显示,TUG1高表达与p53阳性表达呈正相关关系(r=0.204,P＜0.05).结论 长链非编码RNA TUG1在胃腺癌组织中表达升高,且与p53表达呈正相关关系,有可能成为胃腺癌靶向治疗的新靶标.%Objective Toobserve the expression of taurine up-regulated gene 1 (TUG1) in the gastric cancer tissues and to analyze the correlation between TUG1 and p53 protein.Methods In this study, 108 cases of gastric cancer tissues and adjacent normal tissues were collected.P53 expression level was detected by Immunohistochemistry.The real-time PCR was used to detect the mRNA expression of TUG1, and its relationship with clinicopathological features was analyze.ResultsThe mRNA expression of TUG1 in the gastric cancer tissues was significantly higher than that in the adjacent normal tissues (2.36±1.01 vs.0.93±0.54, P＜0.01).TUG1 was correlated with tumor differentiation, TNM staging, lymph node metastasis, and distant metastasis (P＜0.05).TUG1 was highly expressed in gastric cancer that was poorly differentiated, T3-T4stage, with lymph node metastasis and distant metastasis.However, there was no correlation between TUG1 and gender, age, tumor size, vessel invasion, and perineural invasion.The protein expression of P53 in the gastric cancer tissues was higher than that in the normal tissues (61％vs.39％, P＜0.01).Pearson correlation analysis indicated that the high expression of TUG1 was positively correlated with p53 expression (r=0.204, P＜0.05).ConclusionLncRNA TUG1 expression increases in the gastric cancer and is correlated with the expression of p53 protein, which may be a new target for the therapy of gastric cancer.
