β-TCP/rhBMP-2复合物的制备及其诱导成骨活性观察

         

摘要

为了检测β-磷酸三钙/人重组骨形态发生蛋白-2(β-TCP/rhBMP-2)复合物的骨诱导能力,将自制β-TCP/rhBMP-2复合物植入45只小鼠一侧股后肌袋作为实验组,单纯β-TCP植入对侧作为对照组。术后24小时、72小时、1周、2周、4周和8周分别取材作组织学检查;1、2、4周取材的部分标本作碱性磷酸酶(ALP)活性测定。结果发现实验组有大量新生软骨和骨形成,随着时间推移,生成量逐渐增多;对照组无软骨和骨形成;此外,实验组ALP活性明显高于对照组(P<0.001),并且随时间推移而升高。表明β-TCP/rhBMP-2具有良好的骨诱导作用。%To determine the osteoinductive potential of β-TCP/rhBMP-2 composite,the β-TCP/rhBMP-2 composite was obtained by combining Beta-tricalcium phosphate (β-TCP) prepared with recombinant human morphogenetic protein-2 (rhBMP-2)and was implanted into muscle pouches in the thigh of mice. β-TCPalone was implanted on the opposite site as controls.At intervals of 24 and 72 hours and 1,2,4 and 8 weeks after the implantation,the specimens were obtained,and histologic study and alkaline phosphatase assay(1,2,4 weeks)were performed.It was found that there was a large amount of new cartilage and bone formation within the composite,and incereasing with time.Meanwhile,there was no new bone formation where β-TCP alone was implanted.The levels of alkaline phosphatase in the β-TCP/rhBMP-2 implants also were increasing with time and were higher than those in controls (P<0.001).The results suggest that β-TCP/rhBMP-2 possesses heterotopic osteoinductive potential.
